چکیده

The fact that the tissue affinity of thiamine propyl disulfide (TPD) is superior to that of thiamin hydrochloride has been shown by many studies in this country as well as in foreign countries. Especially, the distribution of thiamine in the blood after administration of TPD is worthy of note, i.e., the thiamine concentra tion in the blood, particularly in blood cells, is markedly higher after administra tion of TPD than after administration of thiamine hydrochloride. However, the mechanism of penetration of TPD into blood cells in such high concentrations remained to be elucidated. In order to clarify this, the authors gave TPD-S35 (outer), TPD-S35 (inner) and thiamine-S35 to animals under different conditions, investigated the distribution of both S35 and thiamin and obtained several interesting findings.

Absorption, utilization and clinical effectiveness of allithiamines compared to water-soluble thiamines.

Oral administration of lipid-soluble allithiamines [thiamine propyl disulfide (TPD) and thiamine tetrahydrofurfuryl disulfide (TTHF)] rapidly increased thiamine activity in whole blood, red blood cells, cerebrospinal fluid, and urine in normal and thiamine-deficient subjects. These thiamine congeners also restored red blood cell transketolase to normal in alcoholics with thiamine deficiency. Su...

Inhibition of thiamine transport in Saccharomyces cerevisiae by thiamine disulfides.

Both thiamine disulfide and O-benzoyl thiamine disulfide, which are thiolfrom derivatives of thiamine, strongly inhibited thiamine transport in Saccharomyces cerevisiae. The inhibition appeared to be due to a high affinity of the analogs for yeast cell membranes, in which thiamine transport component(s) may be integrated.

The Journal of Vitaminology 12, 221-230 (1966) Retention in the Body of Various O-acyl Thiamine Disulfides and Several Proper Ties of O-dutyryl-t Iamine Disulfide

In the course of a seires of investigations on TDS derivatives, the authors have hitherto found easily absorbable BIDS, thlaminic acid (an oxidation product of thiamine), and thiamine disulfide sulfmonoxide and their O-aryl derivatives (1-10). In the present paper, studies were made on the relation between the number of carbon atoms of the acyl group and the uptake by erythrocytes, retention in...

Human brain activation in response to olfactory stimulation by intravenous administration of odorants.

To identify the BOLD effects related to olfaction in humans, we recorded functional magnetic resonance imaging (fMRI) scans in response intravenously instilled thiamine propyl disulfide (TPD) and thiamine tetrahydrofurfuryl disulfide monohydrochloride (TTFD). TPD and TTFD evoked a strong and weak odor sensation, respectively. Since we did not spray the odor stimuli directly, this method is expe...

Benfotiamine, a synthetic S-acyl thiamine derivative, has different mechanisms of action and a different pharmacological profile than lipid-soluble thiamine disulfide derivatives

BACKGROUND Lipid-soluble thiamine precursors have a much higher bioavailability than genuine thiamine and therefore are more suitable for therapeutic purposes. Benfotiamine (S-benzoylthiamine O-monophosphate), an amphiphilic S-acyl thiamine derivative, prevents the progression of diabetic complications, probably by increasing tissue levels of thiamine diphosphate and so enhancing transketolase ...
